Feature:
The seed propagation of the monkey-faced orchid (Dracula simia) is extremely difficult, which requires sterile
environment and professional equipment. The following are laboratory-level operation guidelines:
First, seed preparation
1. Seed treatment: the surface of the mature capsule is disinfected with 75% alcohol, and the seeds are peeled
off in a sterile environment.
2. Culture medium: MS medium +0.1% activated carbon +1% sucrose, pH 5.8-6.0.
3. Equipment: clean bench, constant temperature incubator (20-25℃), LED plant fill light.
Second, the sowing process
1. Sterilization: the seeds are soaked in 10% sodium hypochlorite solution for 8 minutes and washed with
sterile water for 3 times.
2. Inoculation: evenly spread on the surface of the culture medium in the ultra-clean table, and seal the culture bottle.
3. Dark culture: keep it completely dark for the first 30 days, with humidity of 80% and temperature of 22℃ 1℃.
4. Illumination stage: after germination, switch to 12 hours/day weak light (2000-3000 lux).
