Sephadex G-10 Medium is well established gel filtration medium for desalting and buffer exchange of biomolecules >700 molecular weight. This is the dry solid form, with particle size range 40 - 120 μm. After full hydration, which takes hours, this material will swell to several mL per gram of powder. Solutes with ≤700 Da molecular weight will be fully or partly included in the hydrated bead volume. While in principle gel filtration size resolution can be achieved, typically one uses G-10 columns for "desalting," buffer exchanges to remove small solutes, since almost all macromolecules are excluded and emerge together from the column.
This bottle contains vintage Sweden-made gel filtration medium repackaged by Aldrich (although the Certificate of Analysis, from April 2003, specifies Supelco, yet another repackaging brand owned by Sigma-Aldrich; you can download it yourself from the Millipore Sigma site by searching for LOT 01424DB). The manufacturer sells 100 g minimum (catalogue number 17-0010-01), but sometimes you do not need that much. Sephadex is made of cross-linked dextran, which doesn't seem to care much about the passing of time, so long as it is unused and dry, as this item is. So while this item has been stored at room temperature for decades, it would probably be difficult to identify any performance difference from Sephadex currently sold by a company that has been renamed a few times. Please do not purchase this (discounted) item if you do not agree.
